چکیده

The activity of alanine-cr-ketoglutarate transaminase (n-alanine :2-oxoglutarate aminotransferase, EC 2.6.1.2) is increased markedly in the livers of diabetic or fasted rats (1). The induction of this enzyme also occurs when either intact or adrenalectomized rats are fed diets high in protein or given corticosteroid injections (24). The adaptive response of this enzyme in glucocorticoid-sensitive tissues, such as thymus (4) and Walker carcinoma 256 (5), is of particular interest since most studies of adaptive enzymes have been restricted to liver. Lin and Knox (6) and Sereni, Kenney, and Kretchmer (7) have shown that tyrosine-cr-ketoglutarate transaminase (n-tyrosine : 2-oxoglutarate aminotransferase, EC 2.6.1.5) is increased several fold in the livers of rats treated by injection with cortisol. Before undertaking studies directed toward an understanding of the mechanism of enzyme induction and the physiological significance of the altered activity of these enzymes, further information on the comparative responses of alanine and tyrosine transaminases seemed desirable. The present report describes a comparative study of these two transaminases with special reference to (a) the time course of the response after the injection of cortisol, (5) induction by substrates, (c) effects of adrenalectomy on the endogenous and cortisol-induced levels, (d) effects of fasting, high protein diets, and alloxan diabetes, and (e) their response in cortisol-sensitive tissues. Differences in the specificity and nature of induction of these two adaptive enzymes were noted. A preliminary report of certain aspects of this work has appeared (8).

Properties of an Aminotransferase of Pea (Pisum sativum L.).

A transaminase (aminotransferase, EC 2.6.1) fraction was partially purified from shoot tips of pea (Pisum sativum L. cv. Alaska) seedlings. With alpha-ketoglutarate as co-substrate, the enzyme transaminated the following aromatic amino acids: d,l-tryptophan, d,l-tyrosine, and d,l-phenylalanine, as well as the following aliphatic amino acids: d,l-alanine, d,l-methionine, and d,l-leucine. Of othe...

Inactivation of bacterial D-amino acid transaminase by beta-chloro-D-alanine.

Purified D-amino acid transaminase from Bacillus sphaericus catalyzes an alpha, beta elimination from the D isomer of beta-chloroalanine to yield equivalent amounts of pyruvate, chloride, and ammonia; the L isomer of chloroalanine is not a substrate for this transaminase. During the beta elimination there is a synchronous loss in enzyme activity; the Kinact for beta-chloroalanine was estimated t...

Regulation of insulin release by factors that also modify glutamate dehydrogenase.

Leucine and monomethyl succinate initiate insulin release, and glutamine potentiates leucine-induced insulin release. Alanine enhances and malate inhibits leucine plus glutamine-induced insulin release. The insulinotropic effect of leucine is at least in part secondary to its ability to activate glutamate oxidation by glutamate dehydrogenase (Sener, A., Malaisse-Lagae, F., and Malaisse, W. J. (...

Factors influencing the development of tyrosine-alpha-ketoglutarate transaminase activity in rat liver.

Previous studies in this laboratory (1, 2) demonstrated that the livers of the fetal rat and of the premature human infant exhibit a markedly reduced ability to oxidize tyrosine, in contrast to the much higher activity of livers obtained from neonatal rats and full term infants. The reduced activity of the livers of premature infants and fetal animals was ascribed to lack of tyrosine-oc-ketoglu...
